色谱类仪器定性不是强项,分离才是它的强项。气相色谱也是如此,但是如果将气相色谱与质谱联用,作成色质联谱,则可以解决其定性能力不强的问题。
但是如果采用TCD、FID、ECD、FPD检测器,则要依靠保留值来定性,比如保留时间、调整保留时间、保留体积、相对保留体积、以及相对保留值。其中相对保留值相对比较好一点,因为不受色谱柱径、柱长、填充情况、流速的影响,只要柱温、固定相一定,相对保留值就一定。
那么依靠保留值定性的依据是什么呢?对于不同模式的气相色谱来说可能不一样,以分配色谱为例,应该是一定温度下组分在固定相和流动相达到平衡时的分配系数或者说分配比。由于不同组分在两相间的分配系数不一样,导致不同组分的迁移速度不同,从而流出色谱柱的时间也不同,当不同组分依次流出色谱柱时,依靠保留值就可以实现定性。
所以有人说:差速迁移是所有色谱类仪器分离的共同原因。
